ARTHROPLASTYMEMBRANE-DERIVEDFIBROBLASTSINDUCEOSTEOLYSISINASEVERECOMBINEDIMMUNODEFICIENTMICEMODELABSTRACTAim:TodevelopaSCID(severecombinedimmunodeficientmice)modelofosteolysisinducedbyArthroplastymembrane-derivedfibroblasts.Methods:theexperimentwascarriedoutintheLaboratoryAnimalCenterofshanghaiSixthPeople’sHospitalfrommatch2006toapril2007.FifteenSCIDmicewereselectandrandomlydividedinto3groups:FibroblastswereisolatedfromperiprosthetictissueduringtherevisionandarticularcapsuleduringtheprimaryTHA.ThenthefibroblastswereimplantedontothecalvarialoftheSCIDmice,whichwasdividedintothreegroups:groupI:Arthroplastymembrane-derivedfibroblastsgroup,groupII:capsulederivedfibroblastsgroupandgroupIII:controlgroup.groupⅠ:Thefibroblastswerereceivedfromperiprosthetictissue，afterpassagingthecells3-4times,thefibroblastswereimplantedontothecalvarialoftheSICDmice,groupⅡ:thefibroblastswerereceivedfromarticularcapsule,groupⅢunderwentshamsurgeryonly.Aftertwoweeks,calvariawereprocessedforhistomorphometryandtheboneresorptionareawascalculatedautomaticallybythesoftware.Result:ThesecellsderivedfromArthroplastymembranewerepositiveforthefibroblast-associatedantigen,prolyl-4-hydroxylase.ExpressionofRANKLandOPGmRNAwerefoundinarthroplastyderivedfibroblastculture.AftertheimplantationofArthroplastymembrane-derivedfibroblasts,theboneresortionwasobservedinthegroupⅠ.Theboneresorptionareainthemidlinesuturewas0.083±0.013mm2incapsulederivedfibroblastsgroup(groupⅡ),0.078±0.009mm2incontrolgroup(groupⅢ),0.23±0.030mm2inArthroplastymembrane-derivedfibroblastsgroup(groupⅠ)(P0.05).Conclusion:themethodofreceivingfibroblastsfromarthroplastymembraneismaneuverable.BoneresorptionwasconfirmaftertheimplantationofArthroplastymembrane-derivedfibroblastsontothecalvarial,thismodeldemonstratesareproduciblemethodforstudyingperiprostheticosteolysisseeninpatientswithasepticloosening.KEYWORD:fibroblasts,arthroplasty,osteolysis,animalmodel目录引言.............................................................................................
