不断增大，CON1 组、CON2 组、CON3 组、DM1 组、DM2 组、DM3 组视网膜 MOD 值分别为0.215±0.019 、 0.225±0.024 、 0.215±0.021 、 0.302±0.031 、 0.523±0.047 、0.748±0.078，且各比较组间差异均有统计学意义（P0.05）；PEDF 阳性染色随病程进 展而减弱，且 MOD 值也不断减小，CON1 组、CON2 组、CON3 组、DM1 组、DM2 组、DM3 组 视网膜 MOD 值分别为 0.908±0.073、0.915±0.031、0.898±0.029、0.735±0.087、0.607±0.055、0.187±0.039，且各比较组间 P0.05。qPCR 示随病程延长，糖尿病组 大鼠视网膜 VEGF mRNA 表达量与正常对照组相比不断增多，CON1 组、CON2 组、CON3 组、 DM1 组、DM2 组、DM3 组 VEGF 表达量分别为 0.105±0.014、0.099±0.129、0.106±0.012、0.372±0.037、1.031±0.095、2.364±0.198，且各比较组间 P0.05；糖尿病组大鼠视 网膜 PEDF mRNA 表达量与正常对照组相比不断减少，CON1 组、CON2 组、CON3 组、DM1 组、 DM2 组、 DM3 组 视 网 膜 PEDF 表 达 量 分 别 为 2.643±0.188 、 2.589±0.136 、 2.533±0.1521.528±0.093 、 0.905.±0.111 、 0.254±0.077 ， 且 各 组 间 P0.05 。Western-blot 示随病程进展，糖尿病组大鼠视网膜 VEGF 蛋白含量增加，PEDF 蛋白含量 减少。CON1 组、CON2 组、CON3 组、DM1 组、DM2 组、DM3 组 VEGF 蛋白表达量分别为 0.328±0.028 、 0.317±0.048 、 0.331±0.035 、 0.436±0.034 、 0.651±0.028 、0.793±0.026，各比较组间 P0.05；PEDF 表达量分别为 0.796±0.063、0.802±0.034、0.799±0.041、0.605±0.086、0.424±0.065、0.263±0.067，且各比较组间 P0.05。结论：1.糖尿病大鼠视网膜细胞超微结构病理改变是最早能观察到的视网膜组织病 变，且其病变程度随病程进展而加重；2.糖尿病大鼠视网膜血管形态结构在糖尿病病程 中、后期开始发生病变，且随病程进展而加重；3. 糖尿病大鼠血-视网膜屏障功能在病 程初期已出现损害，且其严重程度随病程进展而加重；4.视网膜 VEGF、PEDF 表达失衡 在糖尿病初期就已存在，随病程进展，VEGF 表达不断增多，PEDF 表达不断减少，即二 者的失衡存在于糖尿病发展全程。关键词： 糖尿病性视网膜病变，血管上皮生长因子，色素上皮衍生因子The pathological changes of retina of diabetic rats and itsexpression of VEGF and PEDF in different timesABSTRACTObjective: To observe pathologic changes of retina of diabetic rats in different period , and to research the dynamic expression of VEGF and PEDF in retina .
