新疆医科大学医学硕士学位论文StudiesonexpressionandsignificanceofFasandHSP70inbeaglespinalcordafterspineintensitymodulatedradiationtherapyandconventionalradiationtherapyPostgraduate：XUWan-longSupervisor：Prof.BAIJing-pingAbstractObjective:Bycomparingthespineintensitymodulatedradiationtherapy(IMRT)andtheconventionalradiationtherapyofthebeaglespinalcordneuronstodemonstratethebiologicalsafetyofIMRTofthespinalcord.Methods:Selectpurebredbeaglesandsixteenwererandomlydividedintofourgroups.Mimicabeagleclinicalmodeloftumorintheninthandtenththoracicvertebrae.ThenfourgroupsbeaglewereirradiatedbytwodifferentmodelsofIMRTandconventionalradiationtherapy，andthetotalirradiationdoseswere50Gyand70Gyin20days.Thesamplesofspinalcordweretakenoutonthe3thmonthafterradiation.2-DEtechniquewasusedtoidentifythedifferentialproteininthespinalcordafterspineIMRTandconventionalradiationtherapy.Thesampleswereobservedbythehematoxylinandeosinstain.TheexpressionofFas,HSP70andFas,HSP70mRNAwereevaluatedinspinalcordneuronsbyimmunohistochemistryandRT-PCR.TerminaldeoxynucleotidyltransferasemediateddUTPnickandlabeling(TUNEL)techniquewasusedtoexaminetheapoptoticcellsinthespinalcord.Results:①ThedifferentialproteinsofFasandHSP70wereidentifiedbythe2-DEtechniqueinthespinalcordafterspineIMRTandconventionalradiationtherapy.②ComparedwiththeIMRTgroup,thescoreoftheethologyintheconventionalradiationtherapygroupwashigherfollowingthesamedose.③Thelateradiotherapyresponseofthespinalcordneuronswasobviouslyobservedonthe3thmonth.TheneuronsinthespinalcordofIMRTgroupweremainlyreversibleinjury,buttheconventionalradiationtherapyonesweremainlyapoptosis.④ComparedwiththeIMRTgroup,theapoptosisrateofthespinalcordneuronsandtheexpressionofFasandFasmRNAintheconventionalradiationtherapygroupincreasedobviouslyfollowingthesamedose.⑤TheexpressionofHSP70andHSP70mRNAdescended.ThenumerusofFas/HSP70waspositivelycorrelatedwiththeapoptosisrateofthespinalcordneurons.Conclusion:①Thelateradiotherapyresponseofthespinalcordneuronswasexistingonthe3thmonth.②FasandHSP70ta
