LMWH 可以通过下调细胞 VEGF mRNA 的表达水平，抑制肿瘤血管生成，从而 抑制肿瘤转移。 关键词: 低分子肝素；非小细胞肺癌 A549 ；增殖；血管生成 Effects and Mechanisms of Low Molecular Weight Heparin and Cisplatin on Human Lung Cancer Cell Line A549 In Vitro Abstract Objective: To study the effects of LMWH on proliferation, angiogenesis of human lung cancer cell line A549 and its mechanisms. Meanwhile, to investigate possible mechanism of inhibiting cell proliferation 、 influencing cell cycle and inhibiting angiogenesis. And discussed the LMWH and Cisplatin in the mechanism of the changes. Methods: A549 cells were incubated in culture medium in vitro.Effect of LMWH and Cisplatin on the proliferation of A549 cells was measured by MTT colorimetric method. Distribution of cell cycle were examined by flow cytometry. The expression of VEGF gene was analyzed by RT-PCR after treated with LMWH for 24h and 48h. Results: 1 MTT colorimetric method showed that different concentration’s LMWH could inhibit the proliferation of A549 cells. After treated with LMWH for 24h to 48h, compared with control group, the OD values of LMWH-treated groups decreased,and there was statistically significant difference between control group and every treatment group(P0.05). Among every treatment group, there was also statistically significant difference(P0.05). Furthermore, with the increasing concentration of LMWH and prolonging of treatment time, the OD values decreased gradually, that was, LMWH inhibited the proliferation of A549 cells significantly in a dose-and time-dependent manner.
