Methods: The models about the full-thickness cartilage defects are built on the 48 New Zealand white rabbits. The free transplantation of the folded periosteum-bone combined tissue is designed as experiment group. The transplantation of the free periosteum is designed as control group. Meanwhile, the group without any interference is designed as blank group. All rabbits were sacrificed in 2,4,8, and12weeks postoperatively. The conjunction of the repair tissues and perienchyma were examined macroscopically. Haematoxylin—eosin staining and toluidine blue staining were used to observe the characteristics and the thickness of repair tissues. Meanwhile, immunohistochemistry, and scanning electron microscope of were used to evaluated the repair tissue. Histological scores on samples in each group were measured by O＇Driscoll score standard at different time points with light microscope. Results: The repair tissue in experimental group approached to hyaline cartilage, and showed an unclear boundary with the normal cartilage. The repair tissue was dyed deeply with toluidine blue staining. The positive expression of type Ⅱcollagen in the repair tissue was verified. Meanwhile, the capsule of cartilage cells which lining up in order and the porous structure of collagen fibrils were observed by scanning electron microscope. Histological score of the repair tissue was better than in the control group and in the blank group. Conclusion: On the basis of the previous study after
