AB AB STRACT Rheum franzenbachii Munt．is a plant that belongs to the genus Rheum in the family Polygonaceae．Its root and rhizome are used as medicine which is bitter and cold．It has the effect of purgation，purging heat and loosening the bowels and Can be used in the treatment of constipation，jaundice，carbuncle sore，furunculosis，bum and scald，and SO on．In addition，it is antibacterial，antiphlogistic，convergence， hemostatic，etc． Rhaponticin is one kind of the active substances in Rheum franzenbachii．In recent years，researchs have found that rhaponticin has various pharmacological activities，such as decreasing blood-lipid and anti．oxidation．In this paper,using wild Rheum franzenbachii as material s， the study on different types and different concentrations of plant growth regulator in callus induction and proliferation has been conducted，and its effective constituent-rhaponticin was detected by high performance liquid chromatography(HPLC)，which lay a foundation for the further development and utilization in Rheum franzenbachii．The main experimental results are as follows： 1．The leaf of Wild Rheum franzenbachii was used as explamt to induce callus，which was cultured in MS medium supplemented with 6-BA2．0mg／L and different levels of NAA(0．1，0．5，1．0mg／L)．The 3 inductivity inductivity was between 16．7％and 58．3％，and the best formula was medium MS supplemented with 6-BA 2．0mg／L and NAA0．5mg／L． 2．MS medium supplemented with TDZ，KT,6-BA，NAA and 2,4．D alone were all not suitable for call us proliferation．Different levels of 6-BA(0．5，1．0，2．0，4．0mg／L)in combination with NAA(0．5，1．0，2．0mg／L) were beneficial for callus proliferation．6-BA2．0mg／L+NAAl．0mg／L was the best formula for flesh weight proliferation and dry weight proliferation．Fresh weight quantity reached 1 0．1 l g per bottle，and increased 1 9 times；dry weight quantity reached 0．649 per bottle，and increased 1 5 times．This medium is the best one for subculture． 3．In the treatments of different levels of TDZ or 6-BA
