AbstractGastric Abstract Gastric cancer(GC)is leading cause of global cancer mortality responsible for 700000 deaths annuallg and still highly prevalent in many parts of Asia,Eastern Europe and South America．Most patients with GC are diagnosed at advanced disease stages，and overall 5-year survival rates for patients with resectable GC remain low at 1 0-30％despite clinical advances in surgery and therapy．The elucidation of molecules and signaling pathways that drive aggressive and metastatic gastric cancer is likely to offer new avenues for an early indentification and therapeutic intervention of gastric CanCer． Tumor necrosis factor(rNF)一alpha-induced protein 8(TNFAIP8 TIPE) family is recently identified as critical protein for maintaining immune homeostasis and tumorigenesis．The mammalian TNFAIP8 family consists of three members： TNFAIP8，TIPE2，and TIPE3．Among the members，TNFAIP8 promotes tumor growth and metastasis．TIPE2 regulates both carcinogenesis and inflammation，and its germline deletion leads to fatal inflammatory diseases．TIPE3 Can act as a lipid transfer protein and promotes tumorigenesis．But the role of TNFAIP8 family in gastric cancer has only been assessed in a few studies． Our study found that the expression of TNFAIP8 in tumor tissues of gastric cancer was significantly higher than para-carcinoma tissues(P0．05)，TIPE2 and TIPE3 in tumor tissues of gastric cancer Was significantly lower than para-carcinoma tissues(P0．05)．Furthermore，we successfully established human SGC790 1 cell lines and BGC823 cell lines in which TIPE2 Was stably knockdown．We demonstrated that the downregulated expression of TIPE2 promoted cell proliferation stimulated by LPS． Downregulated TIPE2 expression accelerated AKT,Ir,Bot activation and increased S phase proportion in SGC790 1 and BGC823 cells．These results suggest that TNFAIP8 family may be a new potential target for tumor therapy． Keywords：TNFAIP8；TIPE2；gastric cancer II 万方数据 目录中文摘要 目录 中文摘要 I Abstract．．．．．．．．．．．．．．．．．．．．．．．．
