+ The relationship of CD34 、Interleukin-5、Interleukin-5Rα in bone marrow in asthmatic guinea pigs and Arsenic Trioxide’s effection Postgraduate: Jiang Sheng －hua Directed by professor:Liu Xin Abstract Objective: To investigate the relationship of CD34+ 、Interleukin －5、Interleukin － 5Rα in the bone marrow and lung tissue of asthmatic guinea pigs.more importantly ，to study the effects of arsenic trioxide on CD34+ 、Interleukin －5、 Interleukin －5Rα in the bone marrow and lung tissue.and explore the possible therapeutic mechanism of arsenic trioxide of the low dose for asthma from a bone marrow －targeting . Methods : Forty adult male guinea pigs were randomly divided into control group (group A) ， asthmatic group (group B) ， arsenic trioxide group (group C) and dexamethasone group(group D) (ten in each group). Asthmatic model was established by ovalbumin intraperitoneal combined with inhalation . The guinea pigs were killed at 24 h after the last challenge .the total leucocytes and the petcentage of eosinophil in bronchoalveolar lavage fluid (BALF) and bone marrow (BM) were measured. The percentage of hematopoietic CD34+ cells in bone marrow were counted by flow cytometry. Immunohistochemistry detected the expression of CD34+ 、Interleukin －5 、 Interleukin －5Rα in the BM and lung tissue. Interleukin －5 levels were measured by ELISA in BALF and BM.
