208 2007, Vol. 28, No. 05 食品科学 ※生物工程 张莉莉，严群芳，王 恬* (南京农业大学动物科技学院，江苏 南京 210095) ：为制备具有抗氧化活性的大豆生物活性肽，本实验采用酶解处理大豆蛋白粉，对酶解产物进行葡聚糖 Sephadex G-25凝胶柱层析分离，并对其各组分分子量分布及其抗氧化性进行了研究，对抗氧化活性最佳的肽段进 行氨基酸组成分析。结果显示，大豆蛋白酶解物经Sephadex G-25分离后得到了六个肽片段，其中平均链长为4， 即含有2～6 个氨基酸残基的大豆功能短肽SPP4 ，对羟自由基具有最佳的清除作用，清除率达到80.13%，氨基酸 组成分析发现该组分中缬氨酸、亮氨酸、苯丙氨酸、赖氨酸含量较高。结果表明，含2～6 个氨基酸残基的大豆 生理活性短肽具有较好的抗氧化活性。 ：大豆生物活性肽；分子量；羟自由基；抗氧化活性；氨基酸 Study on Isolation and Antioxidant Activity of Soybean Bioactive Peptides ZHANG Li-li，YAN Qun-fang，WANG Tian* (College of Animal Science and Technology , Nanjing Agricultural University, Nanjing 210095, China) Abstract ：To prepare the soybean antioxidant peptides, various peptides in the soybean protein hydrolysates were fractionated by Sephadex G-25 gel filtration, then the scavenging capacity to hydroxyl free radical and the distribution of molecular weights of fractional peptides were determined and at the amino acids compositions of the optimal antioxidative peptide fractions were analyzed. The results showed that six fractional peptides are fractionated by Sephadex G-25 gel filtration from the soybean protein hydrolysates, SPP4 has the maximum scavenging rate to hydroxyl free radical 80.13% and its APCL is 4.The main amino acids in SPP4 are Val, Leu, Phe and Lys. The results indicated that soybean bioactive peptide composed of 2～6 amid acids has strong antioxidant acitivity.
