AbstractAbstract Abstract Abstract Syphilis，a sexually transmitted disease caused by the spirochetal pathogen Treponema pallidum．It has plagued both thepatient and the phy’sician with its protean clinical manifestations．That the immune mechanism of Treponema pallidum is uncertain the diagnostic dilemmas of syphilis．In this study,we analysed the Treponema pallidum-indmed cellular response，and preliminarily discussed the immune mechanism of syphilis．We included 32 syphilis patients and detected the Th cell subsets in these patients．IFN-1，and IL-4 mRNA expression in lesionswere detected by rabbit intracutaneous inoculation．In vivo studys have shown that Thl cells were recruited by Treponema pallidum in peripheral blood and local lesions．We generated peripheral bbod nK)IK)cytes··derived dendritic cellssupplernented with GM·- C SF andⅡ，4 to further analyse the cellar immunity f Treponema pallidum in vitro． We found that dendritic cells be matured by Treponema pallidum．Furthermore， the cell marker CD86 ofdendritic cells was much bwer in syphilis patients than that in healthy donors(PO．01)，and the phagocyticability ofdendritic cells was higher in syphilis patients(P0．01)．We analysed the differentiation of naive Th cells by cocukure with Tp—matured dendritic cells．We found that dendritic cells present Treponema pallidum to naive Th cells．The naive Th cells were actived and prolifemte， expressed surface markers indicative of Thl phemtype，and small number of Treg and Th2 cells at the same time．In this process，the mTOR pathway was actived．It revealed that Treponema pallidum infection actived the imnlune response． In conclusion,Our study indicates that Tp-matured dendritic cells can induce Thl response and activate mTOR pathway．To the best of Our kmwlcdge，this is the first report to analyse the naive Th cell differemiation caused by Treponema pallidum in vitro，which provide experimental basis to T cell and dendritic cell treatment． Keywords：syphilis；naive Th cell；dendritic cell
