摘 摘 要 黄瓜是一种重要的世界性蔬菜，但由于自然侵蚀和品种的杂优化，黄瓜栽培品种的 遗传背景日渐狭窄。因此，开展黄瓜种质遗传多样性的研究，以便从更深入的角度研究 黄瓜种质的遗传分类，为黄瓜育种研究提供选择选配的理论依据。 本研究利用形态学标记和RAPD标记方法对60份不同来源地、不同生态型的黄瓜种 质资源进行了遗传多样性的研究，结果如下： 从120条10碱基随机引物中筛选出20条能产生稳定性、可重复性好的DNA产物的 引物对60份黄瓜种质基因组DNA的扩增结果显示：大多数RAPD标记在黄瓜种质间表现 出多态性。而且，通过严格控制反应体系和反应条件，可以得到重复性好的扩增结果。 因此，通过本实验验证了RAPD标记用于黄瓜种质资源遗传多样性的分析是可行的，可靠 的。 利用形态学标记，50份黄瓜种质被分成4个组群。将不同地域来源的黄瓜种质很好 的区分开，按照形态特征可大体分成华南型和华北型两大类。基于RAPD标记·60份黄 瓜种质共分成8个组群，其中第8组群又可再分成3个亚组。从各组群的地理来源看， 可以将来源不同国家的黄瓜种质区分开。从形态特性看，可以将中国的华南型黄瓜、华 北型黄瓜和欧洲型黄瓜品种基本区分开，与传统的分类方法基本一致。 通过利用两种方法对黄瓜种质分类加深了对种质资源的认识深度，更精确的了解种 质之间的遗传相似度，这为杂交育种亲本的选择选配提供了一定的理论依据。在本研究 的两种分类方法中，本课题组培育的品种631，与其它种质遗传距离较远。而且该品种 商品性和抗病性均显示出很大的优势，因此，用该品种傲亲本与其他品种杂交可能将会 有较大的杂种优势。 关键词黄瓜；种质资源：遗传多样性；形态学标记；RAPD V ，。。。。。一。。，一：，』苎登些型塑圭耋鹜垒圣一。一。。。，。。。一。Study ，。。。。。一。。，一：，』苎登些型塑圭耋鹜垒圣一。一。。。，。。。一。 Study on the Genetic Diversity of Cucumber (Cucumis sativus L．)Germplasm Abstract Cucumber is㈦important worldwide vegetable．The genetic background of cucumber cultivars in production becomes narrower and narrower gradually because of natural genetic erosion and variety hybridization。Therefore，the study on genetic diversity of cucumber accessions will be able to grow on its genetic distribution profoundly and offer the theoretical help to cucumber breeding． In this study,the genetic diversity in cucumber accessions from different countries or regions was assessed by morphological marks and RAPD marks．The results showed： 20 primers producing qualified fragments wero selected from 120 RAPD primers．20 informative RAPD primers were studied among 60 accessions．The results showed：most of RAPD productions were polymorphic．A qualified PCR production could be acquired if we could strictly control the PCR system and program。Therefore，the method of RAPD marker used to assessing the genetic diversity ofcucumber germplasm is feasible and reliable． 50 cucumber accessions were clustered into four groups based on morphological marker． Cucumber accessions from different countries or regions can be
