青海大学医学院 青海大学医学院 2010 级攻读临床医学硕士学位研究生毕业论文 第 第 PAGE 4 页 共 81 页 英文摘要 The Levels of Major Components of the ACE-AngⅡ-AT1 Axis in Local Hematopoietic Bone Morrow in Patients with Chronic Mountain Sickness Postgraduate student: Fu Wei Abstract Background Chronic mountain sickness (CMS) is a clinical syndrome caused by maladaptation to chronic exposure to a high-altitude hypoxic environment and occurs among some individuals inhabiting plateau. CMS is characterized by excessive erythrocytosis and hypoxemia. The pathogenesis of red blood cells excessive accumulation in CMS is still unclear. Researches suggested that there exists an erythropoietin (EPO) independency regulation mechanism. Recent studies indicated the existence of local renin angiotensin system (RAS) in bone marrow haematopoietic tissue. Almost all of RAS components have now been found in the bone marrow hematopoietic microenvironment and hematopoietic cells. Recent data demonstrate that RAS play a role of regulation in the process of physiological and pathological hematopoiesis in an autocrine/paracrine/endocrine fashion. Its regulation function implementation mainly depends on the ACE-AngⅡ-AT1 axis and ACE2-Ang (1-7)-Mas axis. The ACE and AngⅡ in ACE-AngⅡ-AT1 axis contribute to erythroid hematopoiesis. And the AT1 receptors are present on erythroid hematopoietic cells. Hypoxia can induce the gene expression of many local tissues RAS elements such as ACE, AGT and AT1. The basic pathogenesis of CMS is high altitude hypoxia environment, and the outstanding manifestation of CMS is erythrocytosis. Therefor, we assume that the ACE-AngⅡ-AT1 axis of local RAS in bone marrow hematopoietic tissue plays a role in pathological and physiological regulatory mechanism of red blood cells excessive accumulation in CMS.
