ABSTRACT Homogeneous time-resolved fluorescence immunoassay was a kind of time-resolved fluorescence immunoassay which has become a commonly used detection tool in clinical diagnostics, biochemistry and other areas. Those long decay lifetime, large Stokes Shift and sharp emission profile of lanthanide ions chelators made them an ideal choice for use as a tracer. In this dissertation, the following works have been done to meet the needs of this analytical method for organic ligands and chelators: 1. An organic ligand intermediate 6, 6’-dimethyl-[2, 2-bipyridine]-4, 4-dicarboxylate has been synthesized: 2-methyl-6-(tosyloxy) isonicotinate was synthesized via esterification reaction with methyl 2-hydroxy-6-methyl-isonicotinate as a starting material. And targeted product was prepared under the effect of transition metal nickel complex catalysis by homocoupling reaction from 2-methyl-6-(tosyloxy) isonicotinate. The optimum reaction conditions were obtained in 77 % yield by changing the raw material proportion, reflux temperature, reaction time, sequence of adding material. The accuracy of the structure of the product is verified by a variety of characterization methods. 2. A kind of chelator Eu3+-cryptate has been synthesized: The targeted product was synthesized by the raw material diethyl 2,6-dimethylpyridine-3,5-dicarboxylate through NBS bromination, electrophilic substitution and hydrolysis reactions.
