　第17卷　第12期医学研究生学报Vol.17　No.12　2004年12月Journal of Medical P ostgraduates Dec.2004 ?论　　著?去磷酸化RB蛋白在乳腺癌细胞凋亡中的作用 王文武1,　欧阳学农1,　姜　浩2,　曹建国2 (1.南京军区福州总医院肿瘤科,福建福州350025; 2.南华大学附属第一医院肿瘤科,湖南衡阳421001 摘要:　目的:研究去磷酸化R B蛋白表达在人乳腺癌细胞凋亡中的作用。　方法:以乳腺癌细胞株MCF27/S(化疗敏感细胞株和MCF27/Adr(耐多柔比星细胞株为研究对象,采用免疫细胞化学法检测多柔比星(ADR作用前、后R B蛋白的表达状态;应用流式细胞术检测ADR诱导乳腺癌细胞凋亡程度。　结果:加ADR前两种细胞胞核中的磷酸化R B蛋白均为阳性,去磷酸化R B蛋白均为阴性。经不同浓度ADR处理后,随着其浓度的增加,MCF27/S细胞去磷酸化R B蛋白表达量逐渐增高,而MCF27/Adr细胞去磷酸化R B蛋白表达量无明显增高。流式细胞术检测细胞凋亡率和细胞周期,结果表明ADR诱导MCF27/S细胞凋亡作用比MCF27/Adr强(P0.01。　结论:去磷酸化R B 蛋白表达与MCF27/S细胞凋亡呈正相关,而与MCF27/Adr细胞凋亡无明显相关性。 关键词:　去磷酸化R B蛋白;　乳腺癌;　细胞凋亡;　多柔比星 中图分类号:　R737.9　　　文献标识码:　A　　　文章编号:　100828199(20041221059203Ξ The role of dephosphorylated RB protein in human breast cancer cell apoptosis W ANG Wen2wu1,OU2Y ANG Xue2nong1,J I ANG Hao2,C AO Jian2guo2 (11Fuzhou G eneral Hospital o f Nanjing Command,Fuzhou350025,Fujian,China;21the Fir st Affiliated Hospital o f Nanhua Univer sity,Hengyang421001,Hunan,China Abstract:　Objective:T o investigate the role of the expression of dephosphorylated RB protein for cell apop2 tosis in human breast cancer.　Methods:In our w orks,human breast cell lines(MCF27/S,the chem osensi2 tive cell line and MCF27/Adr,the chem oresistent cell linewere evaluated.The expressive levels of dephos2 phorylated RB protein were detected with immunocytochemistry.Apoptosis rates were determined by flow cy2 tometry(FC M.　Results:Before treatment of ADR,phosphorylated RB protein was positive in tw o cell lines,but dephosphorylated RB protein was negative;A fter treatment of different concentration ADR,when the concentration of ADR was increased,expression of dephosphorylated RB protein elevated accordingly in MCF27/S,but no significant change in MCF27/Adr.Apoptosis and cycle was detected by FC M assays shows ADR induced apoptosis of MCF27/S m ore than MCF27/Adr(P0.01.Conclusion:There were positive correlation the expression levels of dephosphorylated RB protein and the apoptosis of MCF27/S cells for Chem osensitivity,but n
