Preparation of Disk-like Natural Hydroxyapatite Samplesand Cell Culture on Their Surfaces

(State Key Laboratory of Bioelectronics, Southeast University, Nanjing, 210096) Abstract: The purpose of this study is to prepare natural hydroxyapatite (NHA) for biocompatibility study. The NHA was prepared from pig bones by means of a calcining procedure, and then were made into disk-like samples. Subsequently mouse mesenchymal stem cells (MSCs) were cultured in vitro and the cell viability metabolism standard curve was obtained. Finally the mouse MSCs were also tried to seed on the surface of disk-like NHA samples? The result showed NHA could be derived from pig bones by a 650°C calcining procedure? MSCs viability metabolism standard curve was obtained and proper cell density of 6000 cells per wells was sclcctcd? The cells grew well on the surface of disk-like NHA samples? These results demonstrated that NHA have good biocompatibility.

0 Introduction
Hydroxyapatite (HA), as an important bone repair material, is generally believed to possess excellent biocompatibility and good osteoconductivity. There are two approaches to obtain HA: chemical synthesis and natural abstraction. Recent studies of HA osteoconductivity are mostly about synthetic HA 12], while few reports are concerning natural HA. Mesenchymal stem cells (MSCs), which have multi-lineage potential, can be induced into osteoblasts on certain conditions. Because of their special differential properties, MSCs are extensively applied in osteoconductivity studies of biomaterials and medicine [3]. This study is designed to prepare disk-like NHA samples for further research of its bone conductivity?

Fresh pig bones were collected for NHA powder preparation.10% polyvinyl alcohol (PVA, AR) solution was prepared
