The oral solution of the complex prescription ganoderma lucidum could prolong significantly prolong the sodium pentobarbitalinduced hou of sleep and increased the incidence of sleep induced by subthreshold dose of sodium pentobarbital in The oral solution of the complex prescription ganoderma lucidum reinforces depth of sleep, improves the sleep quality, and has health care function for sleep improvement. Key words:sleep;Ganoderma lucidum;optimal dose;mice 灵芝（Ganoderma lucidum( Fr.)Ket.)属担子菌亚门，层菌纲，非褶菌目，多孔菌科，灵芝属真菌，俗称灵芝草，主要成分为麦角甾醇、顺蓖麻酸及反丁烯二酸，此外尚含有氨基酸、葡萄糖、多糖类、树脂及甘露醇等［1］.近年来国内外对灵芝的化学成分、药理作用及临床应用等方面进行了广泛的研究.研究结果表明，灵芝具有免疫调节、抗肿瘤、抗病毒、抗衰老、抗疲劳、降血糖、降血压、消炎和镇静催眠等药理作用［2］.复方灵芝口服液是以灵芝为主要原料，配合药食两用的山楂和大枣，经过 科学 的提取及加工制备而成.本研究通过动物实验，探讨复方灵芝口服液改善睡眠功能的最佳配伍剂量. 1 材料与方法 材料 取昆明种小鼠140只，体重为18～22g，由延边大学医学部实验动物科提供.灵芝、大枣及山楂原液由吉林省延边原始林生态保健食品有限公司提供；戊巴比妥钠为上海行知化工厂产品；褪黑素为江苏常州医药技术开发公司产品. 复方灵芝口服液最佳组合剂量选择试验 最佳组合剂量筛选采用L9(34) 3个因素及3个水平设计正交试验，其因素及水平设计见表1.根据表1制备成9种不同配伍比例的复方灵芝溶液（表2）.取90只小鼠，雌雄各半，随机分为9组，即对应复方灵芝的9种剂量组合，每组各为10只.每日灌胃给予小鼠相应复方灵芝溶液1次，剂量为10mL/kg,连续14d.末次灌胃15min后，腹腔注射45mg/kg戊巴比妥钠.以小鼠的翻正反射消失为指标，选择最佳剂量组合［2，3］.表1 因素水平表 表2 复方灵芝剂量组合 复方灵芝口服液改善睡眠的实验 根据正交试验结果,将原液按最佳组合剂量组合后，配制成3种剂量，即高
