甜菜碱对小鼠脾淋巴细胞的增殖作用 季宇彬1,2，冯小燕1,2，高世勇1,2 （1.哈尔滨商业大学生命科学与环境科学研究中心药物研究所博士后科研工作站，哈尔滨150076； 2.国家教育部抗肿瘤天然药物工程研究中心，哈尔滨150076） 摘要：研究甜菜碱对小鼠脾淋巴细胞的增殖作用。应用MTT法观察体外甜菜碱在不同浓度、不同时间对小鼠脾淋巴细胞增殖的影响，应用流式细胞仪（FCM）测小鼠脾淋巴细胞在不同时间周期的变化。结果表明：甜菜碱终浓度0.4mM、4mM、20mM均可促淋巴细胞增值，但终浓度4mM效果最为明显，在甜菜碱刺激24h、48h、72h后，淋巴细胞均可显著增值，但随时间的加倍，增值幅度并不明显，综合考虑，24h效果更好。终浓度4mM甜菜碱淋巴细胞由G0/G1期进入S期，且作用18h效果最明显，其G0/G1期由97.03%下降到89.9%，S期由2.58%升高到9.08%。甜菜碱18h淋巴细胞由G0/G1期进入S期作用效果最Effects of Betaine on Mouse Splen-lymphocytes in Vitro JI yu-bin1,2, FENG xiao-yan1,2, GAO shi-yong1,2 (1. Harbin 150076, China) Abstract: To investigate the effects of Betaine on splen-lymphocytes proliferation, To observe the effects of the Betaine on the lymphocyte proliferation of mouse by MTT at different concentrations and times. To observe the effects of the Betaine on the lymphocyte proliferation cell-cycle of mouse by FCM. The result indicate that Betaine can stimulate lymphocytes proliferation at the concentrations of 0.4mM、4mM and 20mM, but 4mM is the best; Betaine can stimulate lymphocytes proliferation at the time of 24h、48h and 72h, but 24h is the best. Betaine can change the cell-cycle distribution of lymphocytes at different times, and the best time of increasing the proportion of S-phase cells is 18h , raising from 2.58% to 9.08%. So, betaine can stimulate lymphocytes proliferation and the best concentrations is 4mM, the best time is 24h. Betaine can increasing the proportion of S-phase cells, 18h is the best..
