Abstract 0bjective Theaim ofthisstudywastodiscussthetreatmentmechanism oftheprunellaandfritillaria
thunbergiitothyroid—associatedophthahnopathy(TAO)patients． Methods Theorbitaladiposetissuewasobtainedfrom 4
patientswithTAO duringthesurgeryand2normalsubjectsunderwentenucleatedeyesballsduetocornealpenetratinginjury．
Collectedorbita1adiDose tissuewas incubated using explantculture to harvestthe orbitalfibroblasts．Prunella and fritillaria
thunbergiiextract，IFN anddexamethasonewereaddedintomedium respectively．Theproliferation oftheorbitalfibroblastswere
detectedto evaluatethe influenceofdrugsaboveby MTT，and the secretingHA function of cultured orbitalfibroblastswas
assessedbvthehomogeneousmagneticchemiluminescencemethodimmunizations．Theexpressionofadhesionmolecule(ICAM一
1)inthefibroblastswasdetectedbyusingflowcytometry． Results Themigrationandpassagetimeseemedtobesimilar
betweenorhitalfibroblastsfrom TAO patientsandonesfrom normalsubjects．Culturedorbitalfibroblastsshowedthepositive
immunostainingforVimentinandabsentstainingofrDesmin，S-100andCK．Prunellaandfritillaria thunbergiiextractshoweda
similarresuItt0dexamethasonein inhibitingtheproliferationofculturedorbitalfibroblastinTAO patients．Thesameeffectof
prunellaandfritillariathunbergiiextractonthesecretionHA oforbitalfibroblastandexpressingICAM一1inorbitalfibroblastsafter
IFN一 stimulation wasalso seen．Indifferentconcentrationsofprunellagroups，only133mg／mL ofprunellapresented
