Abstract：Thepresentstudyisaimedtoinvestigatethetoxiceffectsofandrographolide (Andro)on
hepatomacellsandelucidateitspreliminarymechanisms． Atiercellsweretreatedwithdifferentconcentrations
ofAndro (0-50 pmolL·一)for24 h，cellviability wasevaluated with 3-(4，5一dimethylthiazol一2一y1)2,5一
diphenyltetrazoliumbromide(MTT)assay．Furthermore，afterhepatomacells(Hep3BandHepG2)weretreated
withdifferentconcentrationsofAndro(0—30pmo1．L )ofr14d，thenumberofcolonyformationwasaccounted
undermicroscope． Cellcyclerelatedproteinssuch asCdc一2，phOsphOrylated-Cdc一2，CyclinB andCyclinD1
weredetectedwithWesternblottingassayandthecellcyclewasanalyzedbyflow cytometryusingpropidium
iodidestaining． MTT resultsshowedthatAndroinduced growthinhibition ofhepatomaceilsinaconcentra—
tion．dependentmannerbuthadno significanteffectson human normalliverL一02 cells． Andro dramatically
decreased the colony formation ofhepatoma cellsin theconcentratiOn—dependentmanner． M oreover．Andro
inducedadecreaseofHep3B cellsattheG0-G1phaseandaconcomitantaccumulationofcellsatG2-M phase．
Atthe molecular leve1．Western blotting results showed thatAndro decreased the expression ofCdc一2．
phosphOrylated—Cdc一2，CyclinD1andCyclinB proteinsinatime—dependentmanner,which raeallcellcycle
