喉癌组织中基质金属蛋白酶MMP┐2及其抑制剂TIMP┐2的表达 【关键词】 喉肿瘤 关键词: 喉肿瘤;基质金属蛋白酶;免疫组化;淋巴转移 摘 要:目的 研究MMP-2及TIMP-2在喉癌组织中表达的情况及其临床相关性. 方法 用免疫组化LSAB法检测64例石蜡包埋的喉鳞癌组织与5例正常喉粘膜组织石蜡标本中MMP-2和TIMP-2的表达. 结果 在淋巴结转移组与非淋巴结转移组MMP-2的阳性表达率分别为74%和39%，TIMP-2的阳性表达分别为52%和24%，经统计学分析二者的χ2 值分别是7.1800（Plt;0.05），5.0420（Plt;0.05），表明明显相关.而MMP-2与TIMP-2的阳性表达率与喉癌的部位、分化程度、临床分级、病理分级均无明显相关. 结论 检测MMP-2和TIMP-2的表达对喉鳞癌的诊断和预后有一定的参考价值. 　　Keywords:laryngeal neoplasms;matrix metalloproteinase;immunohistochemical;lymphatic metastasis 　　Abstract:AIM To study the expressions of matrix metallo-proteinase-2and tissue inhibitor of metalloproteinase-2in hu-man laryngeal squamous cancer and the relationship between the expressions and clinical features.METHODS MMP-2and TIMP-2were examined with immunohistochemical　SAB method in the paraffinembedded specimens from64cases with laryngeal squamous cell carcinoma（LSCC）and5normals who served as the contol.RESULTS In the group of cervical lymphnode involvement and non-cervical lymphn-ode involvement，their positive rates of MMP-2were74%and39%，and their positive rates of TIMP-2were52%and24%，（MMP-2’sχ2 =7.1800，Plt;0.05;TIMP-2’sχ2 =5.0420，Plt;0.05），which showed a significant correlation between them.MMP-2and TIMP-2had no relationship with the situation and malignant degree.Both MMP-2and TIMP-2were expressed more strongly in carcinoma with cervical lymphnode involvement group than in non-cervical lymphn-ode involvement group，but the high TIMP-2expression could not control the activity of MMP-2.Nonbalance be-tween MMP-2and TIMP-2bore no relationship to human la-ryngeal squamous cell carcinoma invasion and metastasis.CONCLUSION The detection of MMP-2and TIMP-2ex-pressions may contribute to diagnosis and prognosis of LSCC. 　　0 引言 　　基质金属蛋白酶（matrix metalloproteinases，MMPs）通过对细胞外基质（extracell matrix，ECM）的降解而促进癌细胞对周围组织的浸润;其抑制剂，基质金属蛋白酶组织抑制因子（tissue inhibitor of metalloteinases，TIMPs）则通过下调金属蛋白酶的活性而抑制肿瘤细胞的侵袭和转移［1，2］ .MMPs及TIMPs在喉鳞癌中表达的研究报道甚少.为了探讨MMPS及TIMPS在喉癌组织
