AbstractConnectivetissuegrowthfactor(CTGF)isamatricellularproteinthatappearstoplayan
importantroleinscarformation.CTGFisamediatorofthepro-fibroticeffectsofTGF-β inculturedcells,but
thespecificfunctionofCTGFinthefibroticprocesshasnotbeenelucidated.ManyactivitiesofCTGFprotein
aremediatedthroughitsdirectbindingtointegrinreceptors.Inthisstudyweinvestigatedtheroleofintegrin
alphavbeta3indifferentiationofhypertrophicscarfibroblasts(HTSF)inducedbyCTGF.First,wecompared
thelevelsofalphav、 beta3integrinsbetweenatrophicscar(AS)andhypertrophicscar(HTS).wefoundthat
alphav、 beta3integrinswereup-regulatedinHTS.Second,weshowthatCTGFinducesHTSFdifferentiation
andcollagensynthesisinmRNAandproteinlevel.Basedontheresults,wedemonstratedtheexpressionof
alphav、 beta3integrinsmRNAwasup-regulatedbyCTGFstimulationduringtheHTSFdifferentiation.Then,
blockadeofthealphavbeta3integrinspreventstheCTGFinducedHTSFdifferentiationandcollagensynthesis.
Furthermore,theincreasedcontractilityoftheHTSFinthecollagenlatticesinducedbyCTGFwasinhibitedby
theIntegrinalphavbeta3blockingantibodies.Inconclusion,ourstudydefinesthealphavbeta3integrinthat
contributestothepro-fibroticCTGFsignalingandsuggeststhatblockadeofthispathwaymaybebeneficialfor
thetreatmentofHTS.
