ImmuneEnhancementofEscherichiacoliHeat-labile Enterotoxiu onNewcastaldiseaseLaSotaStrainVaccine Postgraduate:LiYuehuiAdvisor:SuJingliang Abstract The2recombinantplasmidspLTR72andpLTweretransformedintoE.coliJMI01 andthenculturedat370Cforabout24hoursingrowthmediaLBrespectively.Thewhole celllysatesofalltheseculturedE.coliwerepreparedbythreecyclesofrfozen-thawand sonication60OWXlOsX5untilallwerebrokencompletely.ThemutantsandLTwere expressedrfom thehostofJM101hambouringtheplasmidspLTR72andpLTwere isolatedandpurifiedrfom theirwholecelllysaterespectivelybythemethodof (NH4)2SO4precipitation,Gelflitrationchromatograthy,D-galascosaelution.Thepurified productsinthelysatesweretestedbyGMI-ELISA,theresultsshowedthatthemutants wereexpressedwellwhentheyweretransformedintotheE.cohJMIOIstrainandcultured intheLBmedia.TheresultsofGMI-ELISAshowedthatthemutantshadGMIbinding activiytInaddition,allthepooledsamplesweretested勿SDS,andtheresults showedthatthemutantandLTconsistedoftwosubunits.Themolecularweightwere about11.5KDaforLTBand29KdaforLTA.Afterthesampleswereinjectedintothe lumenoftheligatedRealloopoftherabbitisolatedwithligaturesfor24hours,fluid secretionintothelumenwere5.4mlforI工2.3m1forLTR72and1.6m1forthecontrol.
