Methods Human colon carcinoma cells SW480 were cultured in vitro and treated with 15, 20, 30, 60, 120 and 240 μg/ml of ginsenoside Rh3 for 24, 48 and 72 h, respectively. The proliferation of SW480 cells was determined with MTT colorimetric method. TUNEL and acridine orange/ethidium bromide double staining were used to detect cell apoptosis and calculate apoptosis ratio and necrosis ratio. Results 15 μg/ml of ginsenoside Rh3 treating 48 h was effective to inhibit SW480 cell proliferating, and the effect was enhanced with dose increasing and time prolonging. The inhibition ratio was above 80% when the cells were treated with 30 μg/ml of ginsenoside Rh3 for 72 h. The result of TUNEL showed that apoptotic cells was corrugativus and brow-stained in cytoplasm. The number of apoptotic cell treated for 72 h was higher than that treated for 24 h. The apoptosis ratio was 25.5% of the cells treated with 15 μg/ml of ginsenoside Rh3 for 24 h and 31% with 20 μg/ml of ginsenoside Rh3. The apoptosis ratio was 55%, and the necrosis ratio was 7% of the cells treated with 15 μg/ml of ginsenoside Rh3 for 72 h. The apoptosis ratio was 67.5%, and the necrosis ratio was 7% of the cells treated with 20 μg/ml of ginsenoside Rh3 for 72 h.
