太平洋白蝦免疫機制之研究 The Study of Immunity Mechanism of Pacific White Shrimp,Litopenaeus vannamei 研究生: 蕭雅文 Ya-Wen Hsiao 指導教授:張朴性 Po-Shing Chang 鄭文騰 Win-Ton Cheng [摘要]The purpose of this research was to study the immunity of white shrimp, Litopenaeus vannamei. This study was carried out through three experiments. First part of the experiments was done by using different concentrations of immunostimulants of zymosan, phorbol myristate acetate PMA , laminarin, sodium alginate and trypsin to induce the hemocytes of phenoloxidase PO and superoxidase anion O2- of the shrimps. The results showed that the PO and O2- in the hemocytes of white shrimp were induced some responses when sodium alginate, trypsin, and laminarin were used, whereas the prophenoloxidase activity was inhibited by the soybean trypsin inhibitor STI and benzamidine. The results also showed a prophenoloxidase system do exist in the hemocytes of white shrimp, but it is inactive. It showed that the three concentrations 75 U/μl, 150 U/μl, and 300 U/μl of SOD were effective to clean the O2. in hemocytes of tested shrimp. The second experiment was to test the PO activity, O2-, phagocytosis, clearance efficiency, and total count hemocytes THC were measured at the time intervals of 0, 30, 60, 90, and 120 minutes after using the sodium alginate to induce the factor of immunity and infecting by Vibrio damsela. The results showed that the PO activity and O2- were significantly increased P 0.05 , whereas the phagocytosis and clearance efficiency were significantly decreased P 0.05 .　The THC was increased at first, but decreased afterwards. The third experiment was to test the PO activity, O2-, phagocytosis, clearance efficiency, and total count hemocytes THC in hemocytes of white shrimp, and was measured at the time intervals of 0, 1, 2, and 4 weeks after feeding on algal powders, sodium alginate 500 ppm, 1000 ppm, and 2000 ppm , and an ordinary diet for 30 days.
