断藤益母汤含药血清对类风湿关节炎滑膜成纤维细胞增殖及TNF―α分泌的影响 　　【摘 要】目的：探讨断藤益母汤含药血清对体外培养的类风湿关节炎患者滑膜成纤维细胞增殖以及肿瘤坏死因子-α的影响和机制。方法：体外培养类风湿关节炎滑膜成纤维细胞，取第3～5代细胞，分别加入按临床常规口服剂量折算制取的来氟米特和断藤益母汤家兔含药血清，对照组加入正常家兔血清，MTT法检测24 h、48 h、72 h后各组含药血清对类风湿关节炎滑膜成纤维细胞的影响。ELISA法检测72 h后各组细胞培养上清肿瘤坏死因子-α的含量。结果：相对于对照组，来氟米特和断藤益母汤含药血清作用48 h、72 h，二者均能显著抑制类风湿关节炎滑膜成纤维细胞增殖（Ρ 0.05）；结论：成人临床常规剂量的断藤益母汤含药血清作用48 h、72 h后能显著抑制类风湿关节炎滑膜成纤维细胞增殖，并可降低该细胞肿瘤坏死因子-α分泌量，这可能是该方药改善类风湿关节炎患者临床症状的潜在机制之一。 　　【关键词】 关节炎，类风湿；断藤益母汤；含药血清；滑膜成纤维细胞；肿瘤坏死因子-α；实验研究；动物 　　Effects of Serum Containing Duanteng Yimu Decoction on Proliferation of Fibroblast-like Synoviocytes and TNF-α Production in Patients with Rheumatoid Arthritis 　　Liang Jiang，Lin Chang-song，Lin Yun-bin 　　【ABSTRACT】Objective：To investigate the effects of serum containing duanteng yimu decoction （DYD） on proliferation of fibroblast-like synoviocytes （FLS） and TNF-α （tumor necrosis factor-α，TNF-α） production in rheumatoid arthritis patients.Methods：3 kinds of rabbits serum [normal，serum containing leflunomide （LEF），serum containing DYD] were made.RA-FLS were separated from knee joints of RA patients and subcultured for 3-5 generations.RA-FLS cells were divided into 3 groups： the control group，the LEF group （serum containing LEF） and the DYD group （serum containing DYD） and then proliferation of cells in 3 groups were evaluated by MTT at three time points （24，48，72 h）.After 72 h，cell culture medium was used to detect the TNF-α level with ELISA method.Results：Compared with the control group，after 48 h，72 h，Leflunomide and DYD serums have obviously inhibitive effects on the prolifration of RA-FLS from patients （Ρ 0.05）. 　　Conclusion：DYD can inhibit RA-FLS proliferation in conventional dose and reduce the production of TNF-α，which suggests one of the therapeutic mechanisms of DYD for RA. 　　【Key words】 arthritis，rheumatoid；duanteng yimu decoction；medicated serum；fibroblast-like synoviocytes；tumor necrosis factor-α；experimental study；animal 　　在类风湿关节炎（rheumatoid arthritis，RA）病情进程中，类风湿关节炎滑膜成纤维细胞
