羊肚菌液体菌种培养技术初探 摘要:近年来,我国已进行了羊肚菌培养的研究,但通常是采用固体培养法,对其菌丝球液体培养的研究尚不多见。为此,我们进行了羊肚菌液体菌种培养基配方的研究,以期为羊肚菌菌种的生产提供新的途径。以常规无菌操作技术,将玉米粒大小的菌块接入已灭菌的待用液体培养基中,于24℃静置24h后移至摇床培养箱内震荡培养。对羊肚菌菌丝体液体培养条件的研究表明:当温度为25℃,摇床转速为100r/min,100mL,培养液pH为7.0,可以明显地提高菌丝体的生物量;对羊肚菌进行液体摇瓶培养,以菌丝生物量、菌丝球的大小及个数为筛选指标,通过单因素方差分析,确定了羊肚菌菌丝体生长的最佳液体培养基配方为:马铃薯200g,KH2PO4 2g,MgSO4 0.6g,VB1 0.5mg,1g,1000mL, pH 7.0。 Morchella conica Abstract: In recent years, China has launched a culture of Morchella conica, but they are usually in solid culture, liquid culture mycelium of their research is still rare. Keywords: Morchella conica; Flask culture; Liquid medium 羊肚菌(Morchella conica) 又名美味羊肚菌,俗称羊雀菌、包谷菌等,隶属子囊菌亚门( Ascomycotina) 、盘菌纲( Discomycetes) 、盘菌目( Pezizales) 、羊肚菌科( Morchellaceae) 、羊肚菌属( Morchella) 。据报道,全球羊肚菌属共有30 多个种,分布于亚洲、欧洲、北美洲及大洋洲的一些地区,所以羊肚菌的分布是世界性的。羊肚菌多分布在公路边、河边、山坡、峡谷、火烧林、阔叶林或针阔混交林的腐殖质层土上。生长的土壤类型有黑壤土、山地灰褐森林土、沙壤土、黄壤土等[6]。羊肚菌子实体的生长季节在每年的3 ~5 8~9 , 气温达10 ~20 , 地表温度达12 ~24 , 月降雨量一般35 ~60 mm, 羊肚菌是在低温、高湿条件下萌发子实体的[1]。羊肚菌的菌丝体呈白色, 有分隔, 分隔处有缢缩, 隔膜上有孔洞, 多核, 无锁状联合, 异宗接合, 常产生菌核( 一种坚硬的、无性细胞团, 是羊肚菌的营养贮藏器官, 在休眠阶段可使其渡过不良的气候条件),尚未发现有分生孢子或其他无性孢
