* A more recent cartridge column design that we use at Hewlett-Packard is shown here. This design helps to incorporate in-line sample filtration and clean up devices, like filters and guard columns. A guard cartridge can be easily inserted into the column holders by reversing the collets at the front end. This requires no additional hardware and adds no additional delay volume with respect to the more traditional approach of two columns and tubing. This design also allows for truly resusable end fittings. Since the same column holder will fit any size HP-cartridge column of any of the offered phases, there is no need to remake the end fittings to accommodate a different column. The guards can help to automate some sample prep, and are inexpensive compared to traditional guard columns. * * This slide shows the comparison of a 2 mm id HP cartridge column with and without the integrated guard cartridge. Notice how the run times only shift slightly, by less than +0.5 minutes with the guard. Also, resolution and selectivity are virtually the same between all of the peaks. This design provides an easy to use, automated approach to extending analytical column lifetime without loss to selectivity and resolution. * The outline of this section will be to first cover some troubleshooting of the HPLC system and column problems. We will look at chromatographic baseline symptoms and solutions. We will talk about a troubleshooting guide.
