Vitrobot Operating Instructions
Start up
? Log into FOM and start your session.
? Wear protective cloth, googles, and gloves.
? Open the main valve of the nitrogen gas cylinder. The nitrogen gas is required for operating the plunge
mechanism. Also open the secondary valve on the nitrogen cylinder in case it is closed. The outgoing
pressure should be around 100 PSI.
? Switch on the Vitrobot. The power switch is located at the back side of the device.
? Switch on the monitor.
? Start the VC software by double clicking the VR icon.
? If a pressure error message appears, make sure the main valve and the secondary valve of the nitrogen
gas cylinder are open, and if necessary adjust the pressure with the pressure regulator to around 100
PSI.
? On the monitor select the image with the tweezer in the correct position.
? Place a twisted paper towel on each side of the specimen chamber and soak each with DI water from a
squirt bottle.
? Adjust the parameters for temperature and blotting according to the requirements for your
experiment.
? Click the “Place new Grid” button.
? Place the aluminum “spider”. Fill the Styrofoam cup and the metal cylinder in the center with liquid
nitrogen. Be careful when handling liquid nitrogen and follow the safety standards.
? Top off the liquid nitrogen in the Styrofoam cup. From now on avoid filling liquid nitrogen into the
central metal cylinder.
? After ca. 10 minutes, the central metal cylinder should be cold enough to liquefy ethane gas:
Ethane is flammable, so be careful and make sure there are no sparks or open flames around (e.g.
Bunsen burner). Make sure the secondary valve is closed, then open the main valve of the ethane gas
cylinder. Check the pressure and adjust to around 5 PSI if necessary.
