Methods: B95 8 cell culture supernatant EB virus transformation in hemorrhagic fever with renal syndrome (HFRS patients PBMC, established HFRS patients B LCL, its own B LCL as APC, peptide loading, the stimulation short-term culture G9L specific HFRS patients CD8 + T cells, the application ELISPOT determination of CD8 + T cells by antigenic peptide stimulation produced IFN capabilities. Results: Loading too peptide G9L or V15R’s B LCL can stimulate G9L specific CD8 + T cell activation and produce IFN , but no homologous sequence with the G9L I15P G9L can not stimulate specific CD8 + T cell activation. Conclusion: B LCL can be used as non-professional APC to effectively present the peptide to specific T cells. [Keywords:] B lymphoblast; EB virus transformation; antigen presenting cells; CTL epitope [Abstract] AIM: To establish Epstein Barr virus (EBV) transformed B lymphoblastic cell lines (B LCL) to present peptides as antigen presenting cells (APC) and stimulate short cultured T cells secreting IFN , by which the T cell epitopes are identified . METHODS: PBMCs from patients with hemorrhagic fever with renal syndrome (HFRS) were transformed using EBV from supernatant of B95 8 cells. ELISPOT assay was then employed to evaluate the IFN production of short cultured G9L specific CD8 + T cells stimulated with peptide pulsed autologous B LCL cells. RESULTS: B LCL pulsed with G9L or G9L nested V15R can stimulate G9L specific CD8 + T cells producing IFN , but not B LCL pulsed with non homologous I15P. CONCLUSION: B
