 PAGE \* MERGEFORMAT 15 Effect of enalapril on high glucose-induced mesangial cells to produce TGF1 LN and the expression of CTGF Of: Chen Ping Tang Jing Peng Shuang Cao Wenfu Abstract Objective Enalapril (Enalapril, Ena) on high glucose-induced rat mesangial cells (GMCs) of transforming growth factor 1 (TGF 1), laminin (LN) and connective tissue growth factor (CTGF) expression. The rats GMCs were cultured in normal concentration of glucose (5.5 mmol / L), high glucose (20 mmol / L) and high glucose and different concentrations (10-7mol / L ,10-6mol / L ,10-5mol / L ) in Ena, the same blank control. Were used radioimmunoassay, enzyme-linked immunosorbent assay (ELISA) detection of different time (24,48,72 h) cell culture supernatant of TGF 1, LN content, using reverse transcription polymerase chain reaction (RT PCR) cells detected 72 h the expression of CTGF mRNA. The results compared with the control group, normal glucose group and the high glucose group increased secretion of TGF 1, LN increased synthesis, CTGF expression increased (P lt;0.001), high glucose group was significantly greater (P lt;0.001). Compared with the high sugar, high, medium and low concentrations of Ena intervention can reverse the above changes. Conclusions High glucose can induce the secretion of TGF 1 GMCs increased synthesis of LN increased, CTGF expression increased, Ena reduces extracellular matrix (ECM) accumulation, reduced CTGF expression, thus contributing to delay diabetic glomerulosclerosis (GS) progress .
