VE-cadherin与p120ctn在实验性兔颅内动脉瘤中的表达及其意义

Abstract ABSTRACT Objects: The formation of intracranial aneurysm(IA) was still unclear. In our previous studies, the phosphorylation of VE-Cadherin participated in the progress of formating IA, as well as the translocation of β-catenin into the nucleus of endothelial cells. As a regulating factor of the VE-Cadherin-Catenin compound, p120 catenin(p120ctn) played a critical role in regulating the function of endothelial cells. We designed this study to investigate the hypothesis that VE-Cadherin and p120ctn participate the formation of IA, through building the model of experiment IA in rabbis, and testing the expression of VE-cadherin and p120ctn in IA. Methods: 20 New Zealand rabbits were selected and randomized into 2 groups(experiment group and control, 10 for each). In the experiment group, both carotid arteries of a rabbit were ligated to build the model of experiment IA in rabbits through the operation. In the control group, all rabbits underwent a same procedure as the experiment group , but carotid arteries were not ligated. The speed of blood flow in the basal artery was evaluated by through cranial doppler(TCD) at 1, 7,14, 28,48,56,70,84 days after the procedure accomplished. In the 12th week after the operation, all rabbits were euthanized and the bifurcation of the basal artery were taken and analyzed through: 1, hematoxylin-eosin(HE) staining: morphological changes through the optic microscope. 2, immunofluorescense technique: the expression of VE-Cadherin and P120ctn, the location of P120ctn and its relationship with the core of endothelial cells, Results: 1, the speed
