Acutemyeloidleukemia(AML)arisesfromneoplastictransformationofhematopoieticstemandprogenitorcells,andrelapsed diseaseremainsoneofthegreaterchallengesintreatingthishematologicmalignancy.Thispaperfocusesonangiogenicaspects ofAMLincludingthesigni?canceandprognosticvalueofbonemarrowmicrovesseldensityandcirculatingcytokinelevels.We showthreegeneralmechanismswherebyAMLexploitsangiogenicpathways,includingdirectinductionofangiogenesis,paracrine regulation,andautocrinestimulation.Wealsopresentearlyevidencethatleukemiacellscontributedirectlytovascularendothelia. Noveltreatmentstrategiesareproposed,andareviewofrelevantantiangiogenicclinicaltrialsispresented.Byunderstandinghow bloodvesselscanserveasareservoirforrefractoryandrelapsedAML,newdiagnosticsandpromisingtreatmentstrategiescanbe developed. 1.Introduction Until recently,leukemia studieshavefocusedprimarily on the leukemia cell.
