The water and glycerolchannelaquaglyceroporin-3(AQP3)isabundantinfetalepidermisandmightin?uenceepidermalwaterhandlingand transepidermalwater?uxaroundbirth.ToinvestigatetheroleofAQP3inimmatureskin,wemeasuredinvivotransepidermal watertransportandAQP3expressioninratpupsexposedtoclinicallyrelevant?uidhomeostasisperturbations.Preterm(E18) ratpupswerestudiedafterantenatalcorticosteroidexposure(ANS),andneonatal(P1)ratpupsafteran18hfast.Transepidermal water loss (TEWL) and skin hydration were determined, AQP3 mRNA was quanti?ed by RT-PCR, and in-situ hybridization andimmunocytochemistrywereappliedtomapAQP3expression.ANSresultedinanimprovedskinbarrier(lowerTEWLand skinhydration),whileAQP3mRNAandproteinincreased.Fastingledtolossofbarrierintegrityalongwithanincreaseinskin hydration.ThesealterationswerenotparalleledbyanychangesinAQP3.Toconclude,antenatalcorticosteroids
