Multiple myeloma (MM) is the second most common hematologic malignancy and remains incurable, primarily due to the treatment-refractory/resistantnatureofthedisease.Arationalapproachtothiscompellingchallengeistodevelopnewdrugs thatactsynergisticallywithexistinge?ectiveagents.Thisapproachwillreducedrugconcentrations,avoidtreatmentresistance, andalsoimprovetreatmente?ectivenessbytargetingnewandnonredundantpathwaysinMM.Towardthisgoal,weexamined theantimyelomae?ectsofMAL3-101,amemberofanewclassofnon-ATP-siteinhibitorsoftheheatshockprotein(Hsp)70 molecularchaperone.WediscoveredthatMAL3-101exhibitedantimyelomae?ectsonMMcelllinesinvitro andinvivoina xenograftplasmacytomamodel,aswellasonprimarytumorcellsandbonemarrowendothelialcellsfrommyelomapatients.
