Multiwalledcarbonnanotubeshavebeenshowntopossessunusual?brogenicactivityinvivoandarecurrentlythefocusofintense toxicologicalinvestigations.Thisstudyfurtherdeterminesthe?brogenicpotentialofwell-dispersedMWCNTinhumanlungcell culturemodelsandtodevelopanovelplatformforunderstandingthecellularmechanismsofMWCNT-inducedlung?brosis. Survanta, anatural lung surfactant, showed e?ectiveness in dispersing agglomerates of MWCNT to ?ne structures similarin sizetoaerosolizedone.Atrelevantlowdoses(0.002–0.2μg/cm ),MWCNTexhibitedadose-dependentbio-e?ectonthehuman 2 lungepithelialcellswhichismorepronouncedindispersed-MWCNTcomparedtonon-dispersedform.Signi?cantlyelevated levels of ?brogenic mediators, such as transforming growth factor-β1 and matrix metalloprotienases-9 were observed in the dispersed-MWCNTtreatedlungepithelialcells.Basedonpreviousinvivostudiesshowingthatdispersed-MWCNTpenetrated theinterstitiumandcausedrapidinterstitial?brosis,weevaluatedthepotentialdirectinteractionbetweenlung?broblastsand MWCNT.Directstimulationofhumanlung?broblastcellproliferation,collagenex
