CalciumTriggeredLa-H2PhaseTransitionMonitoredby CombinedRapidMixingandTime-ResolvedSynchrotron SAXS AnanYaghmur*,PeterLaggner,BarbaraSartori,MichaelRappolt InstituteofBiophysicsandNanosystemsResearch(IBN),AustrianAcademyofSciences,Graz,Austria Abstract Background:Awadetal.[1]reportedontheCa2+-inducedtransitionsofdioleoyl-phosphatidylglycerol(DOPG)/monoolein (MO)vesiclestobicontinuouscubicphasesatequilibriumconditions.Inthepresentstudy,thecombinationofrapidmixing andtime-resolvedsynchrotronsmall-angleX-rayscattering(SAXS)wasappliedforthein-situinvestigationsoffaststructural 2+ transitions of diluted DOPG/MO vesicles into well-ordered nanostructures by the addition of low concentrated Ca solutions. Methodology/PrincipalFindings:Understaticconditionsandtheinabsenceofthedivalentcations,theDOPG/MOsystem forms large vesicles composed of weakly correlated bilayers with a d-spacing of ,140A? (La-phase). The utilization of a stopped-flowapparatusallowedmixingtheseDOPG/MOvesicleswithasolutionofCa2+ionswithin10milliseconds(ms).In suchawaythedynamicsofnegativelychargedPGtodivalentcationinteractions,andthekineticsoftheinducedstructural 2+ transitionswerestudied.Ca ionshaveaverystrongimpactonthelipidicnanostructures.Intriguingly,alreadyatlowsalt concentrations (DOPG/Ca2+.2), Ca ions trigger the transformation from bilayers to monolayer nanotubes (inverted 2+ 2+ hexagonalphase,H2).Ourresultsrevealthatabindingratioof1Ca per8DOPGissufficientfortheformationoftheH2 phase.At50uCadirecttransitionfromthevesiclestotheH2phasewasobserved,whereasatambienttemperature(20uC)a shortlivedintermediatephase(possiblythecubicPn3mphase)coexistingwiththeH2phasewasdetected. Conclusions/Significance:ThestrongbindingofthedivalentcationstothenegativelychargedDOPGmoleculesenhances thenegativespontaneouscurvatureofthemonolayersandcausesarapidcollapsingofthevesicles.Therapidlossofthe bilayer stability and the reorganization of the lipid molecules within ms support the argument that the transition
