Wehypothesized thatwomen with aprior single-dose NVP exposure would haveno more thana10% highercumulativeprevalenceoffailureoftheirNNRTI-containingantiretroviraltherapy(ART)overthefirst48wkoftherapy thanwouldwomenwithoutapriorexposure. MethodsandFindings:Weenrolled355NVP-exposedand523NVP-unexposedwomenattwositesinZambia,onesitein Kenya,andtwositesinThailandintoaprospective,non-inferioritycohortstudyandfollowedthemfor48wkonART.Those who died, discontinued NNRTI-containing ART, or had a plasma viral load $400
