ElaD,aDeubiquitinatingProteaseExpressedbyE.coli Andre′ Catic1,2,ShahramMisaghi2¤,GregoryA.Korbel2,HiddeL.Ploegh2* 1ImmunologyProgram,HarvardMedicalSchool,Boston,Massachusetts,UnitedStatesofAmerica,2WhiteheadInstituteforBiomedicalResearchand DepartmentofBiology,MassachusettsInstituteofTechnology,Cambridge,Massachusetts,UnitedStatesofAmerica Background.Ubiquitinandubiquitin-likeproteins(Ubl)aredesignedtomodifypolypeptidesineukaryotes.Covalentbinding ofubiquitinorUblstosubstrateproteinscanbereversedbyspecifichydrolases.Oneparticularsetofcysteineproteases,the CEclan,whichtargetsubiquitinandUbls,hashomologsineukaryotes,prokaryotes,andviruses.Findings.Wehavecloned andanalyzedtheE.coliproteinelaD,whichisdistantlyrelatedtoeukaryoticCEclanmembersoftheULP/SENPproteasefamily thatarespecificforSUMOandNedd8.Previouslymisannotatedasaputativesulfatase/phosphatase,elaDisanefficientand specific deubiquitinating enzyme in vitro. Interestingly, elaD is present in all intestinal pathogenic E. coli strains, but conspicuously absent from extraintestinal pathogenic strains (ExPECs). Further homologs of this protease can be found in Acanthamoeba Polyphaga Mimivirus, and in Alpha-, Beta-and Gammaproteobacteria. Conclusion. The expression of ULP/ SENP-related hydrolasesinbacteriathereforeextendstoplantpathogensandmedicallyrelevantstrainsof Escherichiacoli, Legionella pneumophila, Rickettsiae, Chlamydiae, and Salmonellae, in which the elaD ortholog sseL has recently been identifiedasavirulencefactorwithdeubiquitinatingactivity.Asacounterpoint,ourphylogeneticandfunctionalexamination revealsthatancienteukaryoticULP/SENPproteasesalsohavethepotentialofubiquitin-specifichydrolysis,suggestinganearly commonoriginofthispeptidaseclan.
