The UnfoldedProteinResponse(UPR)isimplicatedinthepathophysiologyofseveralretinaldisordersincludingADRPinP23H Rhorats.TheaimofthisstudywastoexaminetheonsetofUPRgeneexpressioninS334terRhoretinastodetermineifUPR isactivatedinADRPanimalmodelsandtoinvestigatehowtheactivationofUPRmoleculesleadstothefinaldemiseof S334terRhophotoreceptors.RT-PCRwasperformedtoevaluatethegeneexpressionprofilesfortheP10,P12,P15,andP21 stagesofthedevelopmentandprogressionofADRPinS334terRhophotoreceptors.WedeterminedthatduringtheP12– P15period,ERstress-relatedgenesarestronglyupregulatedintransgenicretinas,resultingintheactivationoftheUPRthat wasconfirmedusingwesternblotanalysisandRT-PCR.TheactivationofUPRwasassociatedwiththeincreasedexpression ofJNK,Bik,Bim,Bid,Noxa,andPumagenesandcleavageofcaspase-12thattogetherwithactivatedcalpainspresumably compromise the integrity of the mitochondrial MPTP, leading to the release of pro-apoptotic AIF1 into the cytosol of S334terRhophotoreceptorcells.Therefore,twomajorcross-talkingpathways,theUPRandmitochondrialMPTPoccurin S334ter-4Rhoretinaconcomitantlyandeventuallypromotethedeathofthephotoreceptorcells.
