Methodology/PrincipalFindings:Here,weusedERadeficientmicetoshowthatendothelium-dependentvasorelaxation induced either by red wine polyphenol extract, ProvinolsTM, or delphinidin, an anthocyanin that possesses similar pharmacologicalprofile,ismediatedbyERa.Indeed,ProvinolsTM,delphinidinandERaagonists,17-beta-estradiolandPPT, areabletoinduceendothelialvasodilatationinaortafromERaWild-TypebutnotfromKnock-Outmice,byactivationof nitric oxide (NO) pathway in endothelial cells. Besides, silencing the effects of ERa completely prevented the effects of ProvinolsTManddelphinidintoactivateNOpathway(Src,ERK1/2,eNOS,caveolin-1)leadingtoNOproduction.Furthermore, directinteractionbetweendelphinidinandERaactivatorsiteisdemonstratedusingbothbindingassayanddocking.Most interestingly,theabilityofshorttermoraladministrationofProvinolsTMtodecreaseresponsetoserotoninandtoenhance sensitivityoftheendothelium-dependentrelaxationtoacetylcholine,associatedwithconcomitantincreasedNOproduction anddecreasedsuperoxideanions,wascompletelybluntedinERadeficientmice.
