Mistlberger* DepartmentofPsychology,SimonFraserUniversity,Burnaby,BritishColumbia,Canada Abstract Thedorsomedialhypothalamus(DMH)isasiteofcircadianclockgeneandimmediateearlygeneexpressioninducibleby daytimerestrictedfeedingschedulesthatentrainfoodanticipatorycircadianrhythmsinratsandmice.TheroleoftheDMH intheexpressionofanticipatoryrhythmshasbeenevaluatedusingdifferentlesionmethods.Partiallesionscreatedwiththe neurotoxinibotenicacid(IBO)havebeenreportedtoattenuatefoodanticipatoryrhythms,whilecompletelesionsmade withradiofrequencycurrentleaveanticipatoryrhythmslargelyintact.WetestedahypothesisthattheDMHandfibersof passage spared by IBO lesions play a time-of-day dependent role in the expression of food anticipatory rhythms. Rats received intra-DMH microinjections of IBO and activity and body temperature (Tb) rhythms were recorded by telemetry duringad-libfoodaccess,totalfooddeprivationandscheduledfeeding,withfoodprovidedfor4-h/dayfor20daysinthe middleofthelightperiodandthenfor20dayslateinthedarkperiod.Duringad-libfoodaccess,ratswithDMHlesions exhibitedaloweramplitudeandmeanleveloflight-darkentrainedactivityandTb rhythms.Duringthedaytimefeeding schedule,allratsexhibitedfoodanticipatoryactivityandTbrhythmsthatpersistedduring2dayswithoutfoodinconstant dark.InsomeratswithpartialortotalDMHablation,themagnitudeoftheanticipatoryrhythmwasweakrelativetomost intactrats.Whenmealtimewasshiftedtothelatenight,themagnitudeofthefoodanticipatoryactivityrhythmsinthese caseswasrestoredtolevelscharacteristicofintactrats.Theseresultsconfirmthatratscananticipatescheduleddaytimeor nighttime meals without the DMH. Improved anticipation at night suggests a modulatory role for the DMH in the expressionoffoodanticipatoryactivityrhythmsduringthedailylightperiod,whennocturnalrodentsno
