Genome-WideAnalysisofGLD-1–MediatedmRNA RegulationSuggestsaRoleinmRNAStorage ClaudiaScheckel¤,DimosGaidatzis,JaneE.Wright,RafalCiosk* FriedrichMiescherInstituteforBiomedicalResearch,Basel,Switzerland Abstract TranslationalrepressionisoftenaccompaniedbymRNAdegradation.Incontrast,manymRNAsingermcellsandneuronsare ‘‘stored’’inthecytoplasminarepressedbutstableform.Unlikerepression,thestabilizationofthesemRNAsissurprisinglylittle understood.AkeyplayerinCaenorhabditiselegansgermcelldevelopmentistheSTARdomainproteinGLD-1.Bygenome- wideanalysisofmRNAregulationinthegermline,weobservedthatGLD-1hasawidespreadroleinrepressingtranslationbut, importantly,alsoinstabilizingasub-populationofitsmRNAtargets.Additionally,thesemRNAsappeartobestabilizedbythe DDX6-likeRNAhelicaseCGH-1,whichisaconservedcomponentofgermgranulesandprocessingbodies.BecausemanyGLD- 1andCGH-1stabilizedmRNAsencodefactorsimportantfortheoocyte-to-embryotransition(OET),ourfindingssuggestthat theregulationbyGLD-1andCGH-1servestwopurposes.Firstly,GLD-1–dependentrepressionpreventsprecocioustranslation ofOET–promotingmRNAs.Secondly,GLD-1–andCGH-1–dependentstabilizationensuresthatthesemRNAsaresufficiently abundantforrobusttranslationwhenactivatedduringOET.Intheabsenceofthisprotectivemechanism,theaccumulationof OET–promotingmRNAs,andconsequentlytheoocyte-to-embryotransition,mightbecompromised.
