MathematicalModelofaCellSizeCheckpoint MarcoVilela1,JeffreyJ.Morgan2,PaulA.Lindahl1,3* 1DepartmentofChemistry,TexasAMUniversity,CollegeStation,Texas,UnitedStatesofAmerica,2DepartmentofMathematics,UniversityofHouston,Houston,Texas, UnitedStatesofAmerica,3DepartmentofBiochemistryandBiophysics,TexasAMUniversity,CollegeStation,Texas,UnitedStatesofAmerica Abstract Howcellsregulatetheirsizefromonegenerationtothenexthasremainedanenigmafordecades.Recently,amolecular mechanismthatlinkscellsizeandcellcyclewasproposedinfissionyeast.Thismechanisminvolveschangesinthespatial cellulardistributionoftwoproteins,Pom1andCdr2,asthecellgrows.Pom1inhibitsCdr2whileCdr2promotestheG2RM transition.Cdr2islocalizedinthemiddlecellregion(midcell)whereastheconcentrationofPom1ishighestatthecelltips and declines towards the midcell. In short cells, Pom1 efficiently inhibits Cdr2. However, as cells grow, the Pom1 concentrationatmidcelldecreasessuchthatCdr2becomesactivatedatsomecriticalsize.Inthisstudy,thechemistryof Pom1andCdr2wasmodeledusingadeterministicreaction-diffusion-convection system interactingwithadeterministic model describing microtubule dynamics. Simulations mimicked experimental data from wild-type (WT) fission yeast growingatnormalandreducedrates;theyalsomimickedthebehaviorofaPom1overexpressionmutantandWTyeast exposed to a microtubule depolymerizing drug. A mechanism linking cell size and cell cycle, involving the downstream actionofCdr2onWee1phosphorylation,isproposed.
