MutationoftheZebrafishNucleoporinelysSensitizes TissueProgenitorstoReplicationStress GangaraoDavuluri1.,WeilongGong1.,ShamilaYusuff1.,KristinLorent1,ManimegalaiMuthumani1, AmyC.Dolan1,MichaelPack1,2* 1DepartmentofMedicine,UniversityofPennsylvaniaSchoolofMedicine,Philadelphia,Pennsylvania,UnitedStatesofAmerica,2DepartmentofCellDevelopmental Biology,UniversityofPennsylvaniaSchoolofMedicine,Philadelphia,Pennsylvania,UnitedStatesofAmerica Abstract Therecessivelethalmutationflottelotte(flo)disruptsdevelopmentofthezebrafishdigestivesystemandothertissues.We showthatfloencodestheorthologofMel-28/Elys,ahighlyconservedgenethathasbeenshowntoberequiredfornuclear integrityinwormsandnuclearporecomplex(NPC)assemblyinamphibianandmammaliancells.Maternalelysexpression sustainszebrafishflomutantstolarvalstageswhencellsinproliferativetissuesthatlacknuclearporesundergocellcyclearrest and apoptosis. p53 mutation rescues apoptosis in the flo retina and optic tectum, but not in the intestine, where the checkpointkinaseChk2isactivated.Chk2inhibitionandreplicationstressinducedbyDNAsynthesisinhibitorswerelethalto flolarvae.Bycontrast,flomutantswerenotsensitizedtoagentsthatcauseDNAdoublestrandbreaks,thusshowingthatloss ofElysdisruptsresponsestoselectedreplicationinhibitors.ElysbindsMcm2-7complexesderivedfromXenopuseggextracts. MutationofelysreducedchromatinbindingofMcm2,butnotbindingofMcm3orMcm4intheflointestine.Theseinvivodata indicate a role for Elys in Mcm2-chromatin interactions. Furthermore, they support a recently proposed model in which replicationoriginslicensedbyexcessMcm2-7arerequiredforthesurvivalofhumancellsexposedtoreplicationstress.
