We testedtheeffectofnicotine(akeycomponentofcigarettesmoking)onthetherapeuticeffectsofhumanembryonicstem cell-derivedendothelialcells(hESC-ECs). MethodsandResults:Toinduceendothelialcelldifferentiation,undifferentiatedhESCs(H9line)underwent4-dayfloatingEB formation and 8-day outgrowth differentiation in EGM-2 media. After 12 days, CD31 cells (13.762.5%) were sorted by + FACScanandmaintainedinEGM-2mediaforfurtherdifferentiation.Afterisolation,thesehESC-ECsexpressedendothelial specificmarkerssuchasvWF(96.361.4%),CD31(97.262.5%),andVE-cadherin(93.762.8%),formvascular-likechannels,and incorporatedDiI-labeledacetylatedlow-densitylipoprotein(DiI-Ac-LDL).Afterward,56106hESC-ECstreatedfor24hourswith 28 nicotine(10 M)orPBS(ascontrol)wereinjectedintotheheartsofmiceundergoingLADligationfollowedbyadministration fortwoweeksofvehicleornicotine(100mg/ml)inthedrinkingwater.Surprisingly,bioluminescenceimaging(BLI)showed significant improvement in the survival of transplanted hESC-ECs in the nicotine treated group at 6 weeks.
