Methods:Oralwashsampleswerecollectedfrom20individualsatMemorialSloan-KetteringCancerCenter.16SrRNAgene surveywasperformedby454pyrosequencingoftheV3–V5region(450bp).TargetedidentificationbyDNAmicroarraywas carried out with the Human Oral Microbe Identification Microarray (HOMIM). Correlations and relative abundance were comparedatphylumandgenuslevel,between16SrRNAsequencereadratioandHOMIMhybridizationintensity. Results:Themajorphyla,Firmicutes,Proteobacteria,Bacteroidetes,Actinobacteria,andFusobacteriawereidentifiedwith high correlation by the two methods (r=0.70,0.86). 16S rRNA gene pyrosequencing identified 77 genera and HOMIM identified49,with37generadetectedbybothmethods;morethan98%ofclassifiedbacteriawereassignedinthese37 genera.Concordancebythetwoassays(presence/absence)andcorrelationswerehighforcommongenera(Streptococcus, Veillonella,Leptotrichia,Prevotella,andHaemophilus;Correlation=0.70–0.84).
