OrganicallyModifiedSilicaNanoparticlesAre BiocompatibleandCanBeTargetedtoNeuronsInVivo FardaBarandeh1.,Phuong-LanNguyen1.,RajivKumar2,GaryJ.Iacobucci1,MichelleL.Kuznicki1, AndrewKosterman1,EarlJ.Bergey2,ParasN.Prasad2,ShermaliGunawardena1,2* 1Department of Biological Sciences, The State University of New York at Buffalo, Buffalo, New York, United States of America, 2Institute of Lasers, Photonics and Biophotonics,TheStateUniversityofNewYorkatBuffalo,Buffalo,NewYork,UnitedStatesofAmerica Abstract Theapplicationofnanotechnologyinbiologicalresearchisbeginningtohaveamajorimpactleadingtothedevelopmentof newtypesoftoolsforhumanhealth.Onefocusofnanobiotechnologyisthedevelopmentofnanoparticle-basedformulations foruseindrugorgenedeliverysystems.Howevermostofthenanoprobescurrentlyinusehavevaryinglevelsoftoxicityin cellsorwholeorganismsandthereforearenotsuitableforinvivoapplicationorlong-termuse.Herewetestthepotentialofa novelsilicabasednanoparticle(organicallymodifiedsilica,ORMOSIL)inlivingneuronswithinawholeorganism.Weshowthat feedingORMOSILnanoparticlestoDrosophilahasnoeffectonviability.ORMOSILnanoparticlespenetrateintolivingbrains, neuronalcellbodiesandaxonalprojections.Intheneuronalcellbody,nanoparticlesarepresentinthecytoplasm,butnotin the nucleus.Strikingly,incorporation ofORMOSILnanoparticlesintothebraindidnotinduceaberrant neuronaldeathor interferedwithnormalneuronalprocesses.OurresultsinDrosophilaindicatethatthesenovelsilicabasednanoparticlesare biocompatibleandnottoxictowholeorganisms,andhaspotentialforthedevelopmentoflong-termapplications.
