Egebergetal.Cilia2012,1:15 /content/1/1/15 RESEARCH OpenAccess Primaryciliaandaberrantcellsignalingin epithelialovariancancer DorteLEgeberg1,2,MetteLethan1,RobertManguso1,LindaSchneider1,AashirAwan1,TueSJ?rgensen1, AnneGByskov2,LotteBPedersen1andS?renTChristensen1* Abstract Background:Ovariancanceristhefourthleadingcauseofcancer-relateddeathsamongwomeninDenmark, largelyduetotheadvancedstageatdiagnosisinmostpatients.Approximately90%ofovariancancersoriginate fromthesingle-layeredovariansurfaceepithelium(OSE).Defectsintheprimarycilium,asolitarysensoryorganelle inmostcellstypesincludingOSE,wererecentlyimplicatedintumorigenesis,mainlyduetoderegulationofciliary signalingpathwayssuchasHedgehog(Hh)signaling.However,apossiblelinkbetweenprimaryciliaandepithelial ovariancancerhasnotpreviouslybeeninvestigated. Methods:Thepresenceofprimaryciliawasanalyzedinsectionsoffixedhumanovariantissueaswellasin culturesofnormalhumanovariansurfaceepithelium(OSE)cellsandtwohumanOSE-derivedcancercelllines.We alsousedimmunofluorescencemicroscopy,westernblotting,RT-PCRandsiRNAtoinvestigateciliarysignaling pathwaysinthesecells. Results:Weshowthatovariancancercellsdisplaysignificantlyreducednumbersofprimarycilia.Thereductionin ciliationfrequencyinthesecellswasnotduetoafailuretoentergrowtharrest,andcorrelatedwithpersistent centrosomallocalizationofauroraAkinase(AURA).Further,wedemonstratethatovariancancercellshave deregulatedHhsignalingandplatelet-derivedgrowthfactorreceptoralpha(PDGFRα)expressionandthat promotionofciliaryformation/stabilitybyAURAsiRNAdepletiondecreasesHhsignalinginovariancancercells. Lastly,weshowthatthetumorsuppressorproteinandnegativeregulatorofAURA,checkpointwithforkhead- associatedandringfingerdomains(CHFR),localizestothecentrosome/primaryciliumaxis. Conclusions:OurresultssuggestthatprimaryciliaplayaroleinmaintainingOSEhomeostasisandthatthelow frequencyofprimaryciliaincancerOSEcellsmayresultinpartfromover-expressionofAURA,leadingtoaberrant Hhsignalingandovariantumorigenesis.
