PromiscuousExpressionofH2B-GFPTransgenein HematopoieticStemCells GrantA.Challen1,2,MargaretA.Goodell1* 1CenterForCellandGeneTherapy,BaylorCollegeofMedicine,Houston,Texas,UnitedStatesofAmerica,2MonashUniversity,Clayton,Victoria,Australia Abstract Background:Thestudyofadultstemcellsreliesontheabilitytoisolatethemusingcomplexcombinationsofmarkersfor flowcytometry.Arecentstudyhasusedatetracycline-regulatableH2B-GFPtransgenicmousemodelanalogoustoBrdU pulse-chasemethodstofluorescentlylabelquiescentskinstemcellsinvivo.Inthisstudy,wesoughttousethesemiceto fluorescentlylabelhematopoieticstemcellstostudynicheinteractions. Methods and Findings: We crossed the H2B-GFP mice to mice carrying a tetracycline-regulated transactivator protein. Whenthesemicewereadministereddoxycycline,weobservedagradualdecreaseintotalbonemarrowGFP cellsover + + 12weeks but the hematopoietic stem cell population remained largely GFP (.85%). In histological bone sections, the long-termGFPlabel-retainingcellstendedtoconcentrateattheendostealsurfaceandcompetitivetransplantationassays showedthatthemajorityofhematopoieticstemcellactivitywascontainedintheGFP cellfraction.However,inresponse + to stimulation with 5-fluorouracil, the hematopoietic stem cells of the crossed mice still retained a high level of GFP expressionwhenitwasanticipatedthelabelshouldbelostwhenthecellsdivide.Uponfurtherreview,itwasdetermined thatthefounderH2B-GFPmiceshowedspuriousexpressionofthetransgeneathighlevelsinthehematopoieticstemcell population,thustheobservedresponseofhematopoieticstemcellsinthedoubletransgenicmicetodoxycyclinewasdue toaberrantexpressionofthetransgeneandnotthecorrecttetracycline-regulatablesystem. Conclusions:WeobservedpromiscuousexpressionoftheH2B-GFPtransgeneinthehematopoieticstemcellcompartment ofthebonemarrow.Thisleakyexpressionprohibitstheuseofthismodeltostudyhematopoieticstemcellsinvivoand carefulcharacterizationforeachorganmustbedoneifthistransgenicsystemistobeusedtoisolateotherprospective
