Here, we investigated whether the timing of oligodendrocyte differentiation might be controlled by neuronal differentiation in cerebellar organotypiccultures.Inthesecultures,theslicestakenfromnewbornmiceshowveryfewoligodendrocytesduringthefirst weekofculture(immatureslices)whereastheirnumberincreasesimportantlyduringthesecondweek(matureslices).First, weshowedthatmaturecerebellarslicesortheirconditionedmediastimulatedoligodendrocytedifferentiationinimmature slicesthusdemonstratingtheexistenceofdiffusiblefactorscontrollingoligodendrocytedifferentiation.Usingconditioned mediafromdifferentmodelsofslicecultureinwhichthenumberofPurkinjecellsvariesdrastically,weshowedthatthe effectsofthesedifferentiatingfactorswereproportionaltothenumberofPurkinjecells.Toidentifythesediffusiblefactors, wefirstperformedatranscriptomeanalysiswithanAffymetrixarrayforcerebellarcortexandthenreal-timequantitative PCRonmRNAsextractedfromfluorescentflowcytometrysorted(FACS)PurkinjecellsofL7-GFPtransgenicmiceatdifferent ages.
